Can I delete a video's existing audio track and then add a new audio track?

Generally speaking, sure! 

Just right-click the track in the timeline and choose to separate the audio and video. Then you can delete the audio (or just hide it) and add your new audio!

I delete and replace audios tracks all the time, as after I create the audio track, I export it for volume leveling (using the Levelator) and if necessary, noise reduction (using Audacity). I disable the original audio track while I import and test the modified audio. Once that's done and I'm satisfied with the result, I delete the original audio and drag the new track into place.

Can I do this if someone else created the video? Could I record the screen with audio mute and then add the other audio track?

It's been my own experience that pretty much any MP4 dropped onto the timeline can be separated. But I suppose that could be dependent on how the MP4 was created and Your Mileage May Vary. LOL
